No flushing required. Synthetic oils are fully compatible with conventional oils.
Ford does have a minimum miles before you can switch though, to ensure complete break-in.

For anyone who is interested, a comprehensive tear down and review of various oil filters.
Oil Filters Revealed
The Readers Digest version, with opinions on which filters are recommended, and which to avoid, can be found here:
Oil Filters Revealed - Opinions and Recommendations
Though I remember when they were considered "the best", I would not, and do not, even consider the current generation of Fram filters. They have become cheaply made, slipshod, shadows of their former products relying on what was their "good name" while selling what has now become unreliable junk.
Because the two choices of auto parts stores I have in my small town are an AutoZone, or NAPA, I use NAPA filters which are made by Dana/WIX.

There should be a manual included with this rig, but, if not, download one from this site:
http://owner.ford.com/servlet/ContentServer?pagename=Owner/Page/OwnerGuidePage&year=2006&make=Ford&model=E-450
Nobody that I know has ever gone wrong by following the parts, fluids, and service maintenance intervals recommendations from Ford's engineers. I use Motorcraft 5W20 (syn-blend), the Motorcraft oil filter and air filter, and whatever other fluids and parts are recommended in the manual; my 2003 E450 class C has 72000 trouble-free miles.

FLS 820 and 5
FLS 820 and 5W20 every 3k miles. Yep, I'm old school...every 3k. Engine looks great, runs great and oil looks great @ 35k miles currently.

Synthetic can be introduced at any time, even adding it quart by quart into a dino filled sump.
The real issue is whether you will ever make money using Synth unless you are running extremely cold or hot.
IMHO you never will in normal use.
A quality xW-20 oil that meets specs changed at manufacturer recommended intervals is fine.
As for filters, you can't go wrong with Motorcraft filters.
